when you start a pumping operation and you are using tank water. When the line is stretched and you are told to charge it, does the discharge pressure show you're actual pressure or do you have to wait till the knob is actually open to get an accurate reading from your discharge gauge?

Any line charged, with the valve is fully opened, the line pressure will equal your master discharge pressure.
If a line is charged requiring lower pressure than the master, the valve will NOT be fully opened and the nozzle will have to be opened in order to set the line discharge pressure.
What are your pumping SOPs? THAT will govern your master pressure and how you must charge the lines.
